II. Purification and Properties of Peroxidases from Bamboo Shoot

Two types of peroxidases [EC 1.11.1.7 [EC] ] were obtained from the tissue of bamboo shoot in which the formation of lignin was gradually occurring. One of these peroxidases has absorption maxima at 405 mµ, 498 mµ and 635 rapi in the oxidized form and at 432 mµ and 560 mµ in the reduced form. Based on these spectral properties the enzyme was classified as the A type peroxidase. Oxidation of coniferyl alcohol and guaiacol by the peroxidase in the presence of hydrogen peroxide and the effect of cyanide or azide on the enzyme were studied and the results showed that coniferyl alcohol is a good hydrogen donor for the enzyme.
Another type of peroxidase was obtained in a small amount, and it was classified as the B type peroxidase based on the absorption maxima at 417 mµ and 540 mµ in the oxidized form and at 431 mp, 533 mp and 562 mµ in the reduced form.
The role of peroxidase in lignin formation observed in the tissue of bamboo shoot was discussed.
